Why some times the site of an injection take few days hurting, and some times not

If your arm muscles were relaxed at the time of the injection, you are much less likely to experience soreness after the injection.

Muscle tension is a huge factor, but it also depends on what the injection is, and how beg the needle is. Antibiotics (pennicilin, tetnis) are time release and reside in the area of the shot as a puoch of irritating fluid. Large needles cause your skin to develop a bruise from blood filling the area around the injection (hematoma). Medicines such as pain killers and organ assisting medications move right into your blood stream and reside in individual capilaries, leaving no fluid in the area of the shot.
Several issues. The previous answerer is right: tensing your muscle during the injection will increase the chances of hurting. Also, the volume of fluid delivered makes a difference (the more given, the more discomfort); and how isotonic it is (Plain sterile water hurts like mad; 0.9% saline, which is exactly isotonic, is minimally painful); and most of all, whether or not your body mounts an inflammatory response to it. The inflammatory thing is probably the main variable, to be honest.

Note that it may not be the medication that the reaction is to: it could be to some of the other things that are in the shot, like stabilizers, antibacterials, preservatives and the like that are there to make the stuff stay stable longer on the shelf.

Your vein had an incision from the needle in order for them to draw the blood. Much like a cut on the arm, just on the inside. Like cuts hurt, even small paper cuts, this needs time to heal as well. Hence the pain after a few days. Some time the tolerance of pain is great and you don't feel it as much. Other times you are not so lucky.
I like the first answer. I had to get a tb test and the first time i took it I was nervous and cold and so when she gave it to me, it hurt like a bee sting. But the second tb test didn't hurt at all cuz i was warmer and I wasn't nervouse at all. So just try to remain relaxed the next time you get an injection! *wink*
