Yes, Just like it hurts when you get a shot to numb an area. It has to hurt as it penetrates the skin.
depends on how long it takes the venom to work and the size of the fangs of the spider if the fangs were big enough then yes it would hurt for a second as you were being bitten then the numbness would take over and it would be no big deal.
